Direct Identification of Major Pathogens of the Bubaline Subclinical Mastitis in Egypt using PCR

چکیده
The present study aimed to evaluate the PCR assays (using Universal and specific primers) for detection of the major pathogens of bubaline subclinical mastitis directly from 160 buffaloes milk samples compared to bacterial examination. The bacteriological examinations showed incidence of 23.75%, 15.62%, 3.12% and 0.62% for E. coli, S. aureus, S. agalactiae and S. dysgalactiae respectively. PCR was better in sensitivity and specificity than the conventional culture as it detected more positive results in culturally negative milk samples. The level of sensitivity achieved in our experiments (5x10 1 CFU/ml of milk) is applicable to milk sample analysis without sample enrichment. The results suggest that PCR assay especially the one working with universal primers could be used as an alternative method in routine diagnosis for rapid, sensitive, and specific simultaneous detection of E. coli, S. aureus, S. agalactiae and S. dysgalactiae in milk samples. A polymerase chain reaction based study on the subclinical mastitis caused by Streptococcus agalactiae, S. dysgalactiae and S. uberis in cattle in Ahvaz
Streptococcus agalactiae , S. dysgalactiae and S. uberis are Gram-positive bacterial pathogens that affect cows in dairy herds. These are considered as the major causes of economic losses of dairy producers without a control program. To reveal the role of these agents in inducing subclinical mastitis in dairy cattle in Ahvaz, 100 CMT-positive milk samples and 20 CMT-negative milk samples were c...
متن کاملIdentification of Toxic Shock Syndrome Toxin-1 (TSST-1) gene in Staphylococcus aureus isolated from bovine mastitis milk
Staphylococcus aureus is a major causative pathogen of clinical and subclinical mastitis of dairy domestic ruminants. This agent produces a variety of extracellular toxins and virulence factors including Toxic Shock Syndrome Toxin-1 (TSST-1) which is the major cause of Toxic Shock Syndrome (TSS). In this study 58 S. aureus isolates obtained from 9 dairy herds in East and West Azerbaijan provinc...
متن کاملIsolation, Biochemical and Molecular Identification, and In-Vitro Antimicrobial Resistance Patterns of Bacteria Isolated from Bubaline Subclinical Mastitis in South India
Buffaloes are the second largest source of milk. Mastitis is a major impediment for milk production, but not much information is available about bubaline mastitis, especially subclinical mastitis. The aim of this study was to (a) investigate the application of various tests for the diagnosis of bubaline subclinical mastitis, (b) identify the major bacteria associated with it, and (c) evaluate t...
متن کاملRelationship between in vitro susceptibility of bovine subclinical mastitis isolates and bacteriological outcome of intramammary treatment with cefquinome
The objective of the present study was to determine whether there was an association between the in vitro antimicrobial susceptibility test results of subclinical mastitis pathogens and bacteriological outcomes of intramammary treatments using cefquinome. A total of 110 intramammary pathogens from 51 cows were assessed in this study. Most intramammary infections were due to coagulase-negative s...

Fast and reliable identification of the microorganisms causing mastitis is important for management of the disease and for targeting antimicrobial treatment. Methods based on PCR are being used increasingly in mastitis diagnostics. Comprehensive field comparisons of PCR and traditional milk bacteriology have not been available. The results of a PCR kit capable of detecting 11 important etiologi...
